MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino acid mitochondria-derived peptide (MDP) encoded within the 12S ribosomal RNA gene of the mitochondrial genome. Unlike nuclear-encoded peptides, MOTS-c represents a new class of signaling molecules originating directly from mitochondria, acting as a retrograde signal between mitochondria and the nucleus to regulate metabolic homeostasis, stress response, and cellular longevity.
